Выбор исходного материала и закладка диплоидных инбредных линий в рамках программы гетерозисной селекции картофеля

Аннотация

Актуальность. Современный этап развития генетики и селекции картофеля характеризуется интенсификацией работ ведущих селекционных фирм по внедрению технологии получения диплоидных гибридов F1, размножаемых ботаническими семенами. Однако широкое применение этой технологии осложняется недостатком или отсутствием в международных генетических банках образцов, необходимых для ее инициации. В Институте генетики и цитологии НАН Беларуси создана коллекция селекционного материала для проведения эффективной селекции картофеля на диплоидном уровне. Цель работы – оценить возможность и определить особенности использования этой коллекции для реализации программы маркер-опосредованной селекции диплоидных гибридов картофеля, которая стартовала в Институте генетики и цитологии НАН Беларуси в 2021 году. В задачи исследования входило: оценить эффективность схем получения исходного материала для закладки диплоидных инбредных линий картофеля; отобрать по клубневой продуктивности, признакам культурного картофеля, и устойчивости к болезням и вредителям фертильные самосовместимые диплоидные клоны S1(F2) для последующего создания инбредных линий; получить семена самоопыленных клонов S2(F3). Результаты. Показано, что наиболее эффективными для закладки инбредных линий являются схемы с участием селекционно продвинутых диплоидных клонов на основе межвидовых гибридов и доноров фертильности и самосовместимости Solanum tuberosum L. Всего получено 3212 семян (36,5 семян/ягоду) от самоопыления 38 перспективных гибридов пяти схем. Отобрано для получения инбредных линий самоопыленное потомство 29 образцов диплоидного картофеля. Семена от самоопыления диплоидных клонов обладали пониженной всхожестью и слабым развитием сеянцев: в горшки с торфогрунтом высажено менее 20% возможного их количества. При уборке отобрано по признакам культурного картофеля 167 сеянцев самоопыленных клонов S1. По результатам полевых испытаний 67 клонов S1 из 156 (42,9%) завязали ягоды от самоопыления. Из них отобрали по признакам культурного картофеля и полевой устойчивости к фитофторозу 36 клонов (23,1%). Они ведут происхождение от 17 диплоидных клонов, несущих ДНК-маркеры ряда генов устойчивости к болезням и вредителям. Семена от их самоопыления: всего 4699 шт., из которых 4382 от клонов S1 на основе межвидовых гибридов, будут использованы для получения и отбора самоопыленных клонов S2. Заключение. Использованная в работе коллекция диплоидного картофеля пригодна для закладки инбредных линий. Наиболее эффективные схемы создания исходного материала – на основе межвидовых гибридов и оригинальных линий S. tuberosum доноров фертильности и самосовместимости. Получены семена от самоопыления фертильных самосовместимых клонов S1 на основе исходных диплоидных клонов, несущих ДНК-маркеры ряда ценных для селекции генов устойчивости к болезням и вредителям.
